Spiroazetidines, like all spirocyclic scaffolds, are bestowed with three advantages over their flat counterparts: inherent three dimensional structures may offer more interactions with target proteins; spiroazetidines may provide superior physiochemical properties thus more drug-like; and novel structures may offer fresh intellectual properties. Meanwhile, although no spiroazetidine-containing drugs are current approved for marketing, at least two of them have been advanced to clinical trials. Their applications in medicinal chemistry are destined to grow, especially since many of them are now commercially available.
